Many caterpillars are known for their specific food preferences, which they carry with them as they transform into butterflies. For instance, the monarch butterfly exclusively feeds on milkweed plants, while the Lime butterfly subsists on lime leaves. Despite sharing a common ancestral lineage, these distinct dietary preferences have long been a subject of fascination for researchers.In a previous study conducted by the NUS team, they discovered that when caterpillars were exposed to leaves outside of their usual diet, they would develop a preference for the scent of that new plant after just a few days. Remarkably, these caterpillars then passed on this acquired scent preference to their offspring, a phenomenon that has also been observed in nature when caterpillars encounter a new food plant due to a female butterfly's mistaken egg-laying.

Unraveling the Mechanism: How Caterpillars Inherit Scent Preferences

The researchers were puzzled by how these scent preferences, which originate in the caterpillar's brain, could be communicated to the reproductive cells (egg and sperm) that give rise to the next generation. "Since new food preferences develop in the brain of caterpillars, it was unclear how such preferences were inherited by their offspring," explained Professor Antónia Monteiro, who led the research team.To investigate this intriguing mechanism, the researchers conducted a series of experiments that shed light on the role of the caterpillar's blood, or hemolymph, in facilitating the transfer of learned scent preferences.

The Caterpillar's Blood Holds the Key

The researchers hypothesized that the caterpillar's blood, which bathes both the brain and the reproductive organs, could be the conduit for transporting factors that influence the inheritance of scent preferences. To test this, they conducted a series of experiments:Newly hatched caterpillars were fed either the plants containing the new scent or a control plant. Once the caterpillars matured, their blood was collected and injected into other caterpillars that had not consumed either type of food.The results were remarkable: the caterpillars that received the blood from the control-fed caterpillars maintained their usual food preferences, while those that received blood from the caterpillars fed with the new-scented plants started to lean towards this change in diet. Surprisingly, this preference was also passed on to their offspring, born many days later."This was very surprising to us, as this experiment shows that learning a preference towards a smell can occur without the need for the smell to enter the caterpillar's body via the antennae, as suggested in textbooks," said Prof Monteiro.

These findings suggest a potential mechanism that could facilitate host switching and the formation of new species among caterpillars.
